Pharmacodynamics :
The exact mechanism of action is of the combination is not known. The chemoreceptor trigger zone is stimulated by the neurotransmitters dopamine, histamine and serotonin thereby activating the vomiting centre which causes nausea and vomiting.
Doxylamine Succinate effectively blocks the Chemo Receptor Trigger Zone thereby inhibiting the binding of neurotransmitters and controls Nausea
Pyridoxine Hydrochloride, Exact role unknown. Shown to reduce nausea of pregnancy in many studies
Indications :
Nausea and Vomiting of Pregnancy
Dosage :
1 Tablet OD, at Bedtime
Storage :
Store in cool and dry place, away from light
Pharmacokinetics :
Absorbed mainly from the jejunum (small intestine). Hence enteric-coated tablets used. Peak plasma concentrations, after using delayed-release preparations is around 4-7 hours (Hence the drug is given at bedtime).Doxylamine is metabolised in the liver. Metabolites are excreted through kidneys
Contraindications :
Contraindicated in women with any of the following conditions. Known hypersensitivity to doxylamine succinate, pyridoxine hydrochloride or any inactive ingredient in the formulation. Monoamine oxidase (MAO) inhibitors intensify and prolong the adverse central nervous system effects of anticholinergic (drying) effects of antihistamines
